1. Yes, you can install SetupBuilder on a machine without an active Internet connection. Absolutely no problem. The provided subscription key supports "offline" activation.

2. We make updates available in form of Web Updates ("Live Update") and single-file stand-alone updates. You can download the single-file update from a machine with Internet connection and then copy it to your development machine (e.g. LAN, USB, etc.) to bring your SetupBuilder environment to the latest release.

By the way, I would suggest to disable Web Update checking on this "internet-less" machine (Tools | Options... | Web Update tab -> set "Check for software updates" to "Never").
